醫(yī)學論文范文:甘珀酸對鐵離子誘導的癲癇大鼠縫隙連接及腦電的影響
【摘要】 目的:探究縫隙連接在外傷性癲癇(posttraumatic epilepsy,PTE)發(fā)病機制中的作用。方法:大鼠隨機分為正常組、假手術組、模型組和甘珀酸干預組。觀察大鼠行為學表現(xiàn),并作腦電圖動態(tài)記錄。結(jié)果:大鼠行為學上出現(xiàn)癲癇發(fā)作,腦電圖描記出多種形式的癲癇樣放電波形。與模型組相比,應用甘珀酸干預后,大鼠行為學發(fā)作明顯減少,程度減輕,癇性放電潛伏期不同程度延長,頻率明顯減慢和波幅明顯降低,差別具有統(tǒng)計學意義(P<0.05)。結(jié)論:腹腔注射甘珀酸可明顯減輕癇性發(fā)作,減少癇性放電頻率,降低波幅,提示大量異常的縫隙連接生成可能在PTE形成中起到了重要作用。

The intervention of carbenoxolone to gap junction and the EEG of Fe3+ induced epilepsy in rat

[Abstract] Objective: To explore the role of gap junction in the posttraumatic epilepsy. Methods: Rats were divided into four groups: normal, sham operated, model and carbenoxolone intervention. Behavior of epilepsy was observed and EEG was recorded. Results: Rats displayed typical epileptic behaviors, and epileptic discharges were observed at the same time. Carbenoxolone reduced seizure degree, prolonged the latencies of epileptic discharge, and decreased frequency and amplitude of epileptic waves (P<0.05). Conclusions: Intraperitoneal administration of carbenoxolone could reduce seizure degree. Gap junction might play a role in posttraumatic epilepsy.

外傷性癲癇(posttraumatic epilepsy,PTE)是一種繼發(fā)于腦部創(chuàng)傷后,反復的癇性發(fā)作性疾病,可能出現(xiàn)在頭部外傷后的數(shù)年[1]。它在原發(fā)損傷的基礎上進一步加重了腦組織的病理損傷及神經(jīng)生化改變,惡化病情,增大死亡風險,使臨床處置更趨復雜,給患者帶來了極大的精神負擔和就業(yè)壓力,同時也給家庭和社會帶來了沉重的經(jīng)濟負擔,因此這是一個巨大的醫(yī)學和社會問題。目前對PTE缺乏有效的防治方法,絕大部分患者可發(fā)展為難治性癲癇,其主要原因是對PTE的發(fā)病機制尚未清楚。我們通過建立鐵誘導的大鼠PTE模型,探討縫隙連接(gap junction,GJ)阻斷劑甘珀酸在PTE防治中的應用及相關機制,為GJ阻斷劑用于臨床治療PTE提供必要的信息醫(yī).學.全.在.線52667788.cn。
